Description Trait assayed for Rubisco to total leaf nitrogen content. Total leaf nitrogen
content was determined by CN-Corder (Yanaco Co. Japan) by using the leaf samples
oven-dried at 80C for 3 days. For determining Rubisco content soluble proteins
(2 µg) were subjected to SDS-PAGE using 12.5% (w/v) gels containing 0.1% (w/v)
SDS, and the gels were stained with Coomassie brilliant blue. The intensity of
the band corresponding to Rubisco was measured with an image analyzer (Umax
Technologies, Calif.). The analysis was done on contents derived from leaf
tissue collected 5 days after heading. Phenotypic effect contributed by Kasalath
allele. The means of ten replications per line were used in the data analysis.
Chromosomal locations of putative QTLs were determined by single-factor analysis
(one-way ANOVA) with the general linear model procedure of QGENE. A probability
level of 0.01 was used as the threshold for detecting significant differences in
the mean values of the two genotypic classes, homozygous for Nipponbare and
Kasalath alleles. In the back crossed inbred lines (BILs) there was a high
correlation among the soluble protein content and Rubisco content at 5 days
(r=0.554; P<0.01) after heading
