50. Structure of chloroplast DNA isolated from rice

We isolated rice chloroplast DNA, and constructed a physical map of the DNA (Hirai et al. 1985). The fragments covering the total DNA were also cloned into plasmids. We have determined complete nucleotide sequence of rice chloroplast DNA. The genetic map deduced from the total sequence and a physical map are shown in Fig. 1. The DNA consists of 134,525 nucleotides, which is about 20 kb shorter than tobacco chloroplast DNA. Nevertheless, genes found in tobacco were all found in rice. Details will be reported elsewhere.
