Comments Trait assayed for Rubisco to total soluble protein content. Soluble protein concentration in the supernatant was determined by the method of Bradford with bovine serum albumin as a standard. The protein extract was prepared from leaves by grinding them in liquid nitrogen to a fine powder and suspended in a buffer containing 100 mM NaH2PO4 (pH 7.0), 1 mM phenylmethylsulfonyl fluoride, 1% (w/v) insoluble polyvinyl polypyrrolidone, and 1% (v/v) beta-mercaptoethanol, and then centrifuged at 9,800 g for 5 min. For determining Rubisco content soluble proteins (2 µg) were subjected to SDS-PAGE using 12.5% (w/v) gels containing 0.1% (w/v) SDS, and the gels were stained with Coomassie brilliant blue. The intensity of the band corresponding to Rubisco was measured with an image analyzer (Umax Technologies, Calif.). The means of ten replications per line were used in the data analysis. Phenotypic effect contributed by Nipponbare allele. Chromosomal locations of putative QTLs were determined by single-factor analysis (one-way ANOVA) with the general linear model procedure of QGENE. Two-way ANOVAs for epistatic interaction were also carried out with QGENE at Pd0.01 threshold. The tests were done only with marker loci linked to QTLs with a single-factor P of d0.01 to reduce the likelihood of committing a type-I error.
