Abstract | Ninety three rice varieties from a germplasm collection were screened for the lipoxygenase-3 (LOX-3) content in the embryos using monoclonal antibodies against LOX-3. Sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (PAGE) followed by immunoblotting analysis was used in the qualitative screening procedure. A Thai variety ''Daw Dam'' lacked the LOX-3 band based on the immunoblot analysis. Enzymatic assay for the LOX-3 activity and elution pattern of the LOX-3 activity on DEAE-chromatography confirmed the lack of detectable LOX-3 activity in the seed. These results indicated that the seeds of ''Daw Dam'' lacked LOX-3. |
