Description (aka hcf9) missing PSII thylakoid membrane core complex; green seedling. Green seedling, fluorescence induction kinetics class VII and deficient in thylakoid manganese (Leto and Miles 1977; Miles 1980, 1982; Metz and Miles 1982; Leto et al. 1985). Resistant to diquat (Miles and Leto 1975). Uncovered by TB-1Sb; TB-1Sb-2L4464 (Leto and Miles 1977; Leto 1982; Miles et al. 1985); near npi286 (Burr). Not allelic to hcf1, hcf15, hcf19 (Leto and Miles 1977). W. Cook has now cloned the hcf3 gene from normal maize using the Mu tag. Primary deficiency in photosystem II core complex assembly. Certain PSII polypeptides (32kDa and 47kDa) integrate into thylakoids, then rapidly degrade and at least one PSII polypeptide (51kDa) does not form (Leto and Arntzen 1980; Leto et al. 1985). Small, less-compact grana (Miles and Daniel 1974). Freeze fracture EM shows reduction in large EF particles of grana (Miller 1980; Similar PSII-less phenotype associated with other alleles: hcf3-9, EMS-induced allele (Leto and Miles 1979); hcf3-Mu, Mutator-induced (Cook and Miles 1988). Fast (50-120 ps) fluorescence decay kinetics of thylakoid chlorophyll is not affected by light intensity in hcf3 (Green et al. 1984). Classification: excellent hcf trait. This is perhaps the most striking hcf trait. Difficult to classify as a slightly pale green leaf trait. Viability: lethal at 3-leaf stage. (Polacco). Not allelic to hcf1 or hcf15.
