Description Rice SPK encodes a calmodulin-like domain protein kinase. It is only expressed in the endosperm of immature seeds. Antisense SPK plants lacked the ability to accumulate storage products such as starch, but produced watery seed with a large amount of Suc, as a result of inhibition of Suc degradation. In vitro phosphorylation indicated that SPK phosphorylated specifically a Ser residue in Suc synthase that has been shown to be important for its activity in the degradation of Suc. The watery seed of SPK-deficient mutants contained a decreased amount of proteins and an altered protein composition, in particular, the storage proteins. Lacking of the seed storage substance in spk mutants show no correlation between sucrose concentration in phloem sap and demand for carbon source in the sink organs.
Curator Comment
  • Kawasaki et al. 1993, reported a full-length cDNA encoding SPK was found upstream of the sbe1 gene on chromosome 6.
  • In 1999, Kawasaki et al. isolated rice genomic clones encoding SPK, and found the entire SPK cDNA were located on two different chromosomes. The results of RACE-PCR showed that respective transcripts from SPK-A (on chromosome 6) and SPK-B (on chromosome 10) contained additional sequences which were absent in the SPK cDNA, and these sequences were removed like introns during maturation of the SPK mRNA.
